The emergence of drug resistance in cancer and infectious diseases presents one of the most formidable challenges to modern medicine, undermining the efficacy of established therapies. In this talk, I will present our work on the design and engineering of innovative macromolecules to overcome resistance and enable next-generation targeted gene delivery. We developed antimicrobial polymers that eradicate bacterial and fungal pathogens with high potency, while uniquely preventing resistance even after repeated use. Remarkably, these polymers can also reverse antibiotic resistance phenotypes, thereby restoring the utility of conventional antibiotics. Extending this approach, we engineered anticancer polymers that eliminate tumor cells effectively without inducing resistance, offering a promising new class of therapeutic agents. More recently, we advanced the development of novel lipid nanoparticle (LNP) systems tailored for precise mRNA delivery. These LNPs achieve efficient and tissue-specific transfection while avoiding immunogenicity. They have demonstrated robust efficacy in delivering mRNA vaccines against viral infection and cancer, opening the door to safe, scalable, and durable gene therapies. Together, these innovations illustrate how macromolecular engineering can reimagine therapeutic paradigms, transforming our ability to combat resistance and harness the full potential of gene-based medicine.

楊義燕,新加坡生物工程技術(shù)研究所研究員、新加坡國立大學(xué)醫(yī)學(xué)院研究教授。本科畢業(yè)于合肥工業(yè)大學(xué),博士畢業(yè)于清華大學(xué)。2016年當(dāng)選美國醫(yī)學(xué)與生物工程院(American Institute for Medical and Biological Engineering,AIMBE)院士,2021年當(dāng)選新加坡工程院(Academy of Engineering Singapore, SAEng)院士。現(xiàn)擔(dān)任Nanomedicine: Nanotechnology, Biology and Medicine副主編以及Advanced Healthcare Materials編委。研究方向?yàn)榛诳山到獯蠓肿拥目咕涂鼓[瘤藥物/材料及其遞送系統(tǒng)。在Nature系列(Nat. Mater., Nat. Nanotechnol., Nat. Chem., Nat. Biomedical Eng., Nat. Comms)等權(quán)威期刊發(fā)表論文300余篇,H指數(shù)99,多次入選全球高被引學(xué)者和全球頂尖前10萬科學(xué)家,累計(jì)縱向和橫向合作經(jīng)費(fèi)超過1億元人民幣。曾以第一完成人獲得國家科委科技進(jìn)步二等獎(jiǎng)和國家教委科技進(jìn)步一等獎(jiǎng);曾被評為新加坡“當(dāng)代最有影響力的女性”(Great Women of Our Time);多次入選全球1%高被引學(xué)者;主導(dǎo)研發(fā)的抗菌高分子材料入選《科學(xué)美國人》 “改變世界的十大科技創(chuàng)意”(Top 10 World Changing Ideas),開發(fā)全球首款高分子抗菌噴霧Tirsafe?已在新加坡上市。
